United's youngsters make the trip to Bishop Auckland to face their North east neighbours at Heritage Park (kick-off 7pm GMT) and, with four league fixtures remaining, currently occupy 16th place in the 26-team division.
Diarmuid O'Carroll's side require a top 16 finish to ensure their passage into the play-offs at the end of the 2024/25 campaign and Brayson is keen for United's second-string to build on last week's 4-1 win against ten-man Stoke City, with the 17-year-old striker netting twice against the Potters.
The Walker-born forward, who joined Newcastle's youth set-up in 2015, told newcastleunited.com: "It's a big game because we're looking to get into the top 16 to reach the play-offs. Last week's result was a positive as results haven't been going our way recently so it has given us a big boost.
"Going into the final few games of the season, our main aim is reaching the play-offs so getting back to winning ways helped everyone's confidence going into these matches.
"It is a derby, maybe not as big as when you play Sunderland, but it's still a massive game and one we'll be hoping to get three points from. Every game is huge at this point of the season.
Brayson, son of former Magpies forward Paul, has netted ten goals at under-18 level this term before marking his full home debut for United's under-21 side with a brace and assist against Stoke.
The former George Stephenson High School pupil has been pleased with his form in front of goal during the 2024/25 campaign and, targeting a first professional contract with his boyhood club, is keen to show his worth at under-21 level.
He added: "I thought I played well (against Stoke) but the main thing was the team winning but my two goals have helped towards that so I was so happy to play my part.
"Scoring goals is always big to me. If I play well but miss a big chance, I will think about it so being able to score and help the team feels great.
"The younger players need to step up and show what they've got to help the under-21s and get a place in the team.
"It's what I want to be doing, pushing on and playing in the older age groups to try and get experience to then get into the professional game. It's good to be getting goals and making an impact when I'm playing."
